or after you've already updated to custom firmware?

After - None is needed, you can format your memory stick if you want.

i thought i need a iso folder

and i got some other stuff like seplugins and registry folders and i dont know what they do

Oh thats what you mean. I thought you meant what folders do you still need on your memory stick from the custom firmware update.

Anyways, just format your memory stick and all the folders should automatically be made for you. Remember to back up anything you might want to keep though because when you format the stick it will wipe it clean and install the new files/folders.
